Bad influencers to degrade skin health

    • Lysosome

A lysosome is a membrane-bound cell organelle that contains digestive enzymes. Lysosome plays an important role in cutaneous physiology.
    • Why is it harmful for skin?

UV damages skin cells by lysosomal exocytosis that results in the destruction of the UV protection system in the skin.
    • Reactive oxygen species (ROS)

Reactive oxygen species (ROS) are highly reactive chemical molecules formed due to the electron receptivity of O₂.
    • Why is it harmful for skin?

Increased or prolonged free radical action can overwhelm ROS defense mechanisms, contributing to the development of cutaneous diseases and disorders.
    • Matrix metalloproteinase (MMP)

Matrix metalloproteinases (MMPs) are responsible for degradation of extracellular matrix proteins, which form skin dermal connective tissue.
MMPs are classified as collagenases, gelatinases, stromelysins, and membrane-type MMPs according to their substrate specificities.
    • Why is it harmful for skin?

UV irradiation induces expression of certain members of the MMP family, which degrade collagen and other
extracellular matrix proteins that comprise the dermal connective tissue

Action mechanism of DermaNiA®

DermaNiA® helps to

  • Suppress reactive oxygen species
  • Inhibit oxidative stress (HO-1, SOD1)
  • Inhibit inflammation (NF-κB)
  • Synthesize collagen
  • Decrease MMPs gene expression
  • Inhibit lysosomal exocytosis

Topical solution may not work deep within your cells

Study design

Animal Treatment Duration UV irradiation
6 weeks-old Female SKH-1 hairless mice Oral administration: DermaNiA®, Tricin
Topical application: Retinoic acid
14 weeks 10 weeks

Outcomes

DermaNiA® vs Retinoic acid

UVB irradiation

  • Normal

  • UV

    No treatment
  • Tricin

    Oral administration
  • Retinoic acid

    Topical administration

UVB irradiation

  • Normal

  • UV

    No treatment
  • Tricin

    Oral administration
  • Retinoic acid

    Topical administration
  • Oral administration of DermaNiA® and tricin inhibits wrinkle formation induced by UVB irradiation even more than topical administration of retinoic acid
  • The epidermal thickness decreased by oral administration of DermaNiA® and tricin, but not topical administration of retinoic acid

Collagen biosynthesis

Study design

Animal Treatment Duration UV irradiation
6 weeks-old Female SKH-1 hairless mice Oral administration 10 weeks 10 weeks

Outcomes

Collagen-1 expression

Collagen-1 positive cells (%)

  • The administration of DermaNiA® and Tricin significantly increases Collagen-1 expression and the Collagen-1 positive cells (%) in the dermis.

Collagen fiber (%)

  • The % area of collagen fiber significantly increased in DermaNiA® and Tricin groups compared to UV group.
